悬浮MDCK细胞源与鸡胚源H9亚型禽流感疫苗免疫效力比较研究

摘要:
为探讨MDCK纯悬浮细胞制备的H9亚型禽流感灭活疫苗的免疫效果,使用10日龄SPF鸡胚和MDCK纯悬浮细胞分别接种H9亚型AIV BX13株病毒,收获抗原液,制备2种单价油佐剂灭活疫苗,按照不同免疫剂量对21日龄SPF鸡进行免疫接种,测定免疫后不同时间的血清HI抗体,免疫后21天翅静脉攻毒,测定疫苗的保护率。结果表明:2种疫苗分别以0.3、0.2、0.1、0.02 mL/只的剂量接种21日龄SPF后7 d抗体均为0 log2,免后14 d、21 d时2种疫苗相同免疫剂量组HI抗体无明显规律性差异。免疫后21天,2种疫苗0.1mL/只~0.3mL/只的免疫剂量,各组间HI抗体水平无明显差异,可以达到1:128以上。攻毒试验表明,鸡胚源疫苗0.1 mL/只、MDCK细胞源疫苗0.2mL/只的免疫剂量可以达到10/10保护。本研究为使用MDCK纯悬浮细胞制备的H9亚型禽流感疫苗的深化研究和应用奠定了基础。

Abstract:
In order to study the immune effect of H9 subtype avian influenza inactivated vaccine prepared from MDCK pure suspension cells, the 10 days-old SPF chicken embryos and MDCK pure suspension cells were inoculated with H9 subtype AIV BX13 strain respectively. Two antigen was harvested and with which the two monovalent oil adjuvant inactivated vaccine was prepared. 21 days-old SPF chickens were immunized with different immunization doses of the two H9 subtype avian influenza inactivated vaccines. HI titers were tested at different time after immunization, and the chickens were challenged with the AIV which was the same strain to the vaccine antigen by intravenous route at 21 days after immunization for determination of protection rate. The results showed HI titers of 21 days-old SPF chickens immunized with 0.3 mL、0.2 mL、0.1 mL、0.02 mL/head of the two vaccines were 0 log2 at 7 days after immunization. There was no significant difference in HI titers between the two vaccines in the same dose group at 14 days and 21 days after immunization. At 21 days after immunization, there was no significant difference in HI titers between 0.1 mL/head~0.3 mL/head immune dose groups of the two vaccines,which reached more than 1:128. The challenge test indicated that immune dose of 0.1 mL/head chicken embryo-derived vaccine and 0.2 mL/head cell-derived vaccine reached 10/10 protection. This study laid a foundation for further research and application of H9 subtype avian influenza vaccine prepared by MDCK pure suspension cells.
