HPLC法测定芪蓝囊病饮中的靛玉红

摘要:
建立了芪蓝囊病饮中靛玉红的高效液相色谱(PDA)检查方法。 采用C18(250mm×4.6mm×5μm),柱温35℃,以乙腈-0.2%磷酸水进行梯度洗脱,波长采集范围为200-640nm,流速为1.0mL/min,进样量10μL,提取检测波长为289nm。靛玉红在0.5-2.0μg(R2 =0.9991)线性关系良好,平均加样回收率90.0%(RSD=1.36%,n=6)。所建方法色谱分离较好,前处理简单,可用于芪蓝囊病饮中的靛玉红的含量测定,为完善芪蓝囊病饮的质量标准提供依据。 关键词:靛玉红 芪蓝囊病饮 高效液相色谱法

Abstract:
A method for the determination of indirubin in Qilan Nang disease decoction was developed by the high performance liquid chromatography with photo-diode array detector(HPLC-PDA).It was test with C18(250mm×4.6mm×5μm)column,column temperature 35℃,gradient elution was acetonitrile -0.2% phosphoric acid solution as mobile phase,Photo-diode array detector wavelength range was 200-640 nm,the flow rate was 1.0mL/min,and the injection volume was 10 μL,the recorded wave length was 289nm.In the range of 0.5~2.0 μg/mL of indirubin,the linear relationship is good(R2 = 0.9991),the average recovery was 90.0% with RSD of 1.36%(n=6). The test method has good chromatographic separation and simple pretreatment.It can be used for the determination of indirubin in Qilan Nang disease decoction,which provides a reference for improving the quality standard of Qilan Nang disease decoction .
