猪ST细胞染色体制备及核型分析

摘要:本试验运用空气干燥法制备了猪ST细胞不同代次染色体标本,并对猪ST细胞基础细胞库细胞(63代)、最高限制代次(80代)细胞及高于最高代次10代(90代)细胞进行了核型比较分析,分析结果表明:猪ST细胞染色体数目2n=38。并且对其进行了着丝粒定位,试验结果表明:1~4号为亚中央着丝粒染色体,5~6为亚端着丝粒染色体,7~12为中央着丝粒染色体,13~18为端着丝粒染色体,一对性染色体。在基础细胞库中存在的染色体标志,在最高代次细胞中也存在,与基础细胞库细胞相比,所有细胞的染色体模式数均低于10%,并且核型相同。

Abstract:
Abstract: Different generation swine ST cell chromosome samples were prepared by using air drying technique, and karyotype analysis was carried out among swine ST foundation cell bank (the 63rd generation), the maximum limit generation cells (the 80th generation), and cells 10 generations higher than the maximum limit generation (the 90th generation). The result shows that the number of swine ST chromosome was 2n=38. The positioning of centromere was that: No. 1~4 was submetacentric chromosome, No. 5~6 was elocentric chromosome, No. 7~12 was metacentric chromosome, No. 13~18 was telocentric chromosome and a pair of sex chromosome. chromosome models in foundation cell bank also lie in maximum limit generation cells, and compared to foundation cell bank, the chromosome pattern number of other cells was lower than 10% and the karyotype was same.
