超级细菌NDM-1质粒的水平转移研究

摘要:
为探究超级细菌NDM-1质粒水平转移的可能性,以携带NDM-1质粒的猫源洛菲不动杆菌和人源醋酸钙不动杆菌为供体菌,以工程菌株大肠杆菌DH5α和大肠杆菌、沙门氏菌弱毒菌株为受体菌,通过接合转导试验验证了NDM-1基因通过质粒跨种传播的可能性。结果显示,猫源NDM-1质粒可以转入E. coli DH5α中,并可以DH5α为中介,转入猪霍乱沙门氏菌C500中;而人源NDM-1质粒仅转入E. coli DH5α中,未能转入其它菌株。NDM-1质粒在沙门氏菌C500中比在E. coli DH5α中较为稳定遗传。质粒的导入并不影响宿主菌的生长特性。结果表明NDM-1质粒能够跨越菌种界限,并可以大肠杆菌DH5α为中介,转移至其它菌株中。本研究初步揭示了NDM-1的传播方式。

Horizontal transferability of NDM-1 plasmid

Abstract:
Acinetobacter lwoffii of cat origin and Acinetobacter calcoaceticus of human origin were used as donor strains, Escherichia coli strain DH5α and four attenuated strain(two E. coli strains and two Salmonella strains)were used as recipient strains, and conjugation experiment was conducted to explore the cross-species transferability of NDM-1 mediated by plasmids. Some characteristics of the transconjugants were studied. The NDM-1 plasmid of cat origin was transferred into DH5α, and then to Salmonella choleraesuis C500 strain mediated by DH5α. NDM-1 plasmid of human origin was transferred to DH5α, but failed to transfer to other strains. Furthermore, NDM-1 plasmids in C500 could be more stably inherited than in E. coli DH5α. The plasmid transferred didn’t influence the growth properties of the host strains. The results indicated that NDM-1 plasmids were able to cross the genetic boundaries and transfer to other strains mediated by E. coli DH5α. This study may shed some light on the horizontal transmission of NDM-1.
