In 2023, goslings from a goose farm in Heze city Shandong province experienced symptoms of soft feet, paralysis, and mental paralysis. To determine the cause of the disease in the goose farm, this study conducted autopsy and laboratory diagnosis on the dead goslings sent for examination. This study isolated pathogens from the liver, spleen, brain, and other tissues of infected geese in the farm, and identified them through pathological dissection, RT-PCR detection, virus isolation and genetic evolution analysis. The results showed that a Tembusu virus (TMUV, SDHZ-G strain) was isolated from the goose farm. The SDHZ-G strain was highly virulent and lethal to duck embryos, and ELD50 was 10-3.6/0.2mL. The nucleotide similarity of the E gene was from 89.9% to 99.7%, and the amino acid similarity was from 95.6% to 99.8%, without significant genetic variation. This isolate belongs to the genotype 2.2 clade and is the main endemic clade in Shandong. This study identified the TMUV was the pathogen that causing the disease in the goose farm, and analyzed the genetic evolution characteristics of the pathogen TMUV, providing a reference for the prevention and treatment of gosling diseases in this farm. |
