三种恩诺沙星注射液在猪体内的药物代谢动力学研究

摘要:
三组猪(n=5)分别按2.5mg/kg的剂量肌肉注射拜有利5%、恩诺沙星注射液A、恩诺沙星注射液B三种药物,HPLC法测定血药浓度,winnonlin6.1拟合血药浓度—时间数据,计算参数。三种注射液均符合一室开放模型,峰浓度分别为:0.87μg/mL、0.63μg/mL、0.49μg/mL,AUC分别为6.32 h•μg•mL-1、5.31 h•μg•mL-1、3.45 h•μg•mL-1。实验结果表明:拜有利5%在猪体内的药代动力学参数优于注射液B,注射液B的优于注射液A,结果提示恩诺沙星注射液B与拜有利5%相比临床防治效果略差,药物吸收可做进一步改进。

Abstract:
Three groups of swine(n=5) were injected intramuscularly Baytril 5%, enrofloxacin injectionA, and enrofloxacin iniejction B. Plasma concentration was measured by HPLC, and concentration-time data was processed by winnonlin6.1. The results showed that models of all groups were best fit by one-compartment open model with peak concentration of 0.87μg/mL, 0.63μg/mL,0.49μg/mL, and AUC of 6.32 h•μg•mL-1, 5.31 h•μg•mL-1, 3.45 h•μg•mL-1. Baytil 5% is the best in pharmacokinetics, then injection B, injection A last. It indicated that injection B was improved significantly, is slightly inferior to Baytril 5%. Its pharmacokinetic attribute of absorption should be improved.
