添加前缀对重组蛋白生物学特性影响的预测分析

摘要:利用分子生物学软件DNASTAR对含有亲和标记、Xa factor识别序列和连接肽的DAB389(Gly4Ser)2-aMSH以及DAB389-aMSH的表位、亲水性以及等电点等生物学特性进行了预测分析,结果显示DAB389(Gly4Ser)2-aMSH的表位为0.8,DAB389-aMSH为2.1;亲水性为2.7高于DAB389-aMSH的1.96;两者的等电点均为5.9;His×Tag、Xa factor识别序列使重组蛋白的亲水性增强,表位降低;重组蛋白氨基端前70个氨基酸中不存在信号肽;DAB389(Gly4Ser)2-aMSH的表达量低于DAB389-aMSH 2%左右,前缀对蛋白表达量有一定的影响。

Abstract:
Abstract: The designed fusion protein DAB389(Gly4Ser)2-aMSH and DAB389-aMSH include affinity labeling, Xa factor, (Gly4Ser)2, was analyzed and compared in some molecular biology characteristics by using molecular biology software DNASTAR. The results showed that the epitope of DAB389(Gly4Ser)2-aMSH was 0.8 lower/less than 2.1 of DAB389-aMSH, it's hydrophilicity was 2.7 higher than 1.96 of DAB389-aMSH, The isoelectric point of them was 5.9. His×Tag, Xa factor recognition sequence made the hydrophilicity strengthen, epitope and antigenicity depress; the pro 70 amino acid in N-terminal of recombination protein had no signal peptide, the expression quantity of DAB389(Gly4Ser)2-aMSH was less 2% than that of DAB389-aMSH, these prefixing had determinate influence to expression protein.
