牛蒡苷元的提取及纯化

摘要:
探索了一种从牛蒡子中提取、纯化牛蒡苷元的方法。将脱脂牛蒡子粗粉用含5%盐酸的50%乙醇溶液回流5 h,过滤后,药渣用50%乙醇溶液重复提取1 h,合并滤液后用5 mol/L NaOH溶液调节至中性,氯仿萃取2次,减压旋转蒸干后得牛蒡苷元粗品。粗品用硅胶柱层析纯化2次,获得类白色粉末,HPLC法进行分析。结果表明,牛蒡苷元样品保留时间与对照品保留时间一致,约为12.1 min,其相对百分含量为99.3%。牛蒡苷元总提取率为1.1%。该方法是一种有效的、简便的、能满足常规实验室设备要求的牛蒡苷元制备方法。

The Extraction and Purification of Arctigenin

Abstract:
The extraction and purification of arctigenin from Fructus arctii was researched in this article. A mixture of degreased fruit powder of Fructus arctii and 50% alcohol solution containing 5% hydrochloric acid was refluxed for 5 hours. After filtration, the solid residue was resuspended in 50% alcohol solution, and refluxed for 1 hour again. The combined filtrate was basified (pH 7) with 5 mol/L NaOH solution and then extracted with chloroform twice. Further, the combined organic phase was evaporated using a rotary evaporator under reduced pressure. The crude extract was purified by silica gel column chromatography twice, and finally obtained the off-white powder. The purified product was analyzed by HPLC. The retention time of the main peak of purified sample was accordant with the time of reference substance of arctigenin,which was about 12.1 minutes. The percent content of arctigenin relative to reference substance was about 99.3%, and the total yield was approximately 1.1%. The results showed that this method was simple, exercisable and suitable for most regular laboratories.
