重组人γ干扰素摇瓶生产工艺优化

摘要:
通过研究不同类型培养基和培养温度、起始pH值等发酵条件对hIFN-γ产量的影响,初步确认了摇瓶水平发酵各因素的最优条件,优化的发酵条件为发酵起始pH值7.0、培养温度37℃、IPTG诱导浓度1.0mmol/L、菌体生长密度OD600达到1.0时加入IPTG、诱导时间为3h。此时hIFN-γ产量达到最高水平,从而为hIFN-γ批量生产奠定了良好的基础。

Optimization of Fermentation Condition of Recombinant Strain BL21(DE3)(pET32a( )-hIFN-γ) in Shaking Flask

Abstract:
The expression level of hIFN-γ was influenced by the different medium and other fermentation conditions including culture temperature, initial pH and so on.Various factors optimal conditions was confirmed by flask- fermentation.The optimal fermentation condition with IPTG induction is culture medium pH7.0,culture temperature 37℃, joining IPTG to final concentration 1.0mmol/L when the recombinant strain growth density OD600 achieved 1.0, and induction time 3h. The production of hIFN-γ reached the highest level.This fermentation technology established a good foundation for the hIFN-γmass production.
