重组人γ-干扰素基因工程菌发酵工艺优化

摘要:
研究不同类型培养基、诱导时间、菌体密度和不同补料对hIFN-γ表达的影响,并运用高密度发酵工艺对hIFN-γ的产量进行优化,目的蛋白表达量从4000mg/L提高到20000mg/L以上,发酵产量提高了5倍,大大提高了效率。同时为验证工艺的稳定性,将优化的发酵条件进行连续三批重复发酵实验。结果表明该优化条件具有重复性,表达稳定,维持在40%左右,从而为hIFN-γ大规模生产奠定了良好的基础。

Optimization of Fermentation Technology of Recombinant Human Interferon-γ

Abstract:
The expression level of hIFN-γ was influenced by the different medium, induction time, cell density and feeding batch. The production of hIFN-γ was optimized by high density fermentation. The expression level of hIFN-γ was increased from 4000mg/L to over 20000mg/L, the fermentation yield was increased by 5 times. In order to confirm the technical stability, we performed a three-time repeated fermentation. The result indicated that the optimal conditions had the repeatability and the expression level remained 40% or so. This fermentation technology established a good foundation for the hIFN-γ mass production.
