HPLC法测定徐长卿中绿原酸的含量

摘要:建立HPLC法测定徐长卿中绿原酸的含量。采用索氏提取法以70%的甲醇水溶液为提取剂对徐长卿中的绿原酸进行提取。利用SHIMADZU C18色谱柱(250mm?4.6mm,5?m),流动相乙腈-0.5%磷酸水溶液(体积比1:9);流速: 1.0mL?min-1;柱温:25℃;检测波长:327nm的条件进行检测。结果表明绿原酸在0.232μg~4.640μg范围内与峰面积呈良好线性关系, r=0.9997。平均回收率为99.32%,RSD为0.216%(n=5)。表明该方法简便、可靠、重现性好,可作为徐长卿中绿原酸含量测定的方法。

Abstract:
Abstract : To establish for the determination the content of Chlomgenic acid in Cynanchum paniculatum (Bge.) Kitag..Using 70% methyl alcohol to extract Chlomgenic acid from Cynanchum paniculatum (Bge.) Kitag by Soxhlet extraction method. Methods with a HPLC method: The determination is measured by HPLC with SHIMADZU-C18(250mm ? 4.6mm ,5?m)column,Using acetonitrile - 0.5% H3 PO4(1:9) as mobile phase.The flow rate is 1.0mL?min-1,detection wave length is 327nm and Column temperature is 25 ℃. The results show that the linear range of chlomgenic acid is 0.232μg~4.640μg,r=0.9997.The average recovery is 99.32% with RSD 0.216%(n=5). The method is simple and reliable with a good reproducibility which could be used to determine chlorogenic acid contents of Cynanchum paniculatum (Bge.) Kitag..
