利用响应面分析法优化鼠李糖乳杆菌产胞外多糖的发酵工艺

摘要:
首先利用Plackett-Burman法筛选出影响鼠李糖乳杆菌胞外多糖产量的三个重要因素,分别为培养温度、酵母粉、接种量,再用最陡爬坡试验逼近产胞外多糖的最大响应区域,最后运用Box-Behnken中心组合方法进行三因素三水平试验,以胞外多糖产量为响应值建立回归模型,采用响应面分析法确定最优工艺参数。结果表明:当温度为29.7 ℃、酵母粉30.45 g/L、接种量5.76%时胞外多糖产量最大,为354.97 mg/L。

Application of response surface methodology for optimization of exopolysaccharide Production by L.rhamnosus

Abstract:
Three important factors (temperature, yeast powder concentration and inoculation volume) related to the production of exopolysaccharide (EPS) were identified by the Plackett-Burman design. Then the steep ascent assay was performed to approach to the biggest response region of parameters affecting EPS production. Subsequently, a regression model for the yield of EPS was established using Box-Behnken central composite design involving the above three factors at three levels. The established model was analyzed by response surface analysis to obtain the optimum fermentation conditions. The optimum temperature, yeast powder concentration, and inoculation volume for EPS production were 29.7℃, 30.45 g/L, and 5.76%, respectively, with a maximum yield of 354.97 mg/L under these conditions.
