脱氧核酶在病毒学中的研究和应用

摘要:
脱氧核酶(DNAzyme)是近年来利用体外分子进化技术筛选得到的一种具有RNA裂解活性的DNA分子,具有核酸酶活性、过氧化物酶活性、激酶活性等多种催化功能及高效性和高特异性,可能成为治疗肿瘤、病毒感染等新型的基因治疗工具。现就DNAzyme的筛选方法、特性和病毒学研究中的应用等方面做一综述。

Abstract:
Deoxyribozmes (DNAzymes) are a set of single stranded DNA which derived from a combinational library of sequence by molecular evolution technique in vitro. They have high catalytical activity and sequence specificity against the target RNA, they can be designed to cleave any target RNA in a sequence specific manner. DNAzymes can inhibit gene expression at mRNA level and thus regulate the expression of corresponding proteins. For this reason DNAzymes may be expected to be a new gene therapeutical tool for malignant tumors, some viral infection diseases and some genetic diseases.
