甜地丁中槲皮素的薄层鉴别和含量测定

摘要:
为完善甜地丁质量标准,采用薄层色谱法对甜地丁中槲皮素的鉴别进行了研究,并用高效液相色谱法测定其含量。薄层色谱以甲苯-乙酸乙酯-甲酸(7∶5∶0.5)为展开剂,高效液相法色谱柱为C18(4.6 mm×250 mm,5 μm),流动相为甲醇-0.3%磷酸溶液(50∶50),检测波长为360 nm,流速1 mL/min,柱温25 ℃,进样量10 μL。结果表明薄层色谱法能对槲皮素实现有效分离,高效液相色谱法槲皮素进样浓度在5~100 μg/mL范围内与峰面积呈良好的线性关系(r=0.9997);样品平均回收率为96.1%(n=6),RSD为0.7%。本方法为进一步控制甜地丁的质量提供了依据。

Abstract:
In order to improve the quality standard of Herba Gueldenstaedtiae ,an TLC method for identification of quercetin in Herba Gueldenstaedtiae was developed. And it’s content was determination by HPLC.The TLC developing agent was methylbenzene – elthlacetate - methanoic acid(7∶5∶0.5).The HPLC conditions was use C18 column(4.6 mm×250 mm,5 μm),mobile phase was methanol - 0.3% phosphoric acid solution (50∶50),detection wavelength was 360 nm, flow rate was 1 mL/min ,column temperature was 25 ℃,the injection volume was 10 μL. Quercetin was effectively speparated by TLC,and there was a good linear correlation between the peak areas and the concentrations of quercetin at the range of 5~100 μg/mL(r=0.9997)by HPLC.The average recovery was 96.1% with RSD of 0.7%(n=6). It can be used for the quality control of quercetin in Herba Gueldenstaedtiae.
