猪多杀性巴氏杆菌病活疫苗(EO630株)生产用合成培养基的研究

摘要:
为便于猪多杀性巴氏杆菌病活疫苗(EO630株)质量控制和降低生产成本,研制了ZJ-1和ZJ-2两种合成培养基,并进行了两种合成培养基与马丁肉汤的比较试验。结果表明,合成培养基ZJ-1培养活菌数高于ZJ-2和马丁肉汤。用合成培养基ZJ-1培养制备3批疫苗,并分别用兔进行安全检验和小鼠效力检验。3批疫苗对兔均2/2健活,对小鼠的保护力分别为9/10、9/10、8/10,符合产品质量标准。本试验表明,运用合成培养基替代传统培养基是可行的。

Abstract:
To facilitate the production of swine Pasteurella multocida vaccine,live(strain EO630) and decrease the manufacturing cost of the culture media, we have developed such two synthetic mediums as ZJ-1and ZJ-2 and made the contrast experiment with Martin broth for effects. The results showed that the amounts of live bacteria prepared by ZJ-1 were significantly higher than those prepared by ZJ-2 and Martin broth. Using ZJ-1 to produce swine Pasteurella multocida vaccine,live(strain EO630), We have got the products in three lots and made related security detection and potency test. In consequence, the results showed that the survival rate of two rabbits were all 100%(2/2) and the protective efficacy for mice were 90%(9/10),90%(9/10) and 80%(8/10) respectively, which met the standard requirements of the quality of products. The experiment suggested that it was feasible to replace traditional culture medium with synthetic medium.
