一株猪肺炎支原体的分离鉴定

摘要:
从全国部分猪场采集到疑似猪支原体肺炎肺组织病料12份,按照猪肺炎支原体PCR法提取DNA进行PCR和多重PCR反应,将检测仅有猪肺炎支原体感染的病料采用剪碎法进行猪肺炎支原体野毒株的分离、传代,最终分离到1株疑似猪肺炎支原体;通过测序分析、形态观察、生化试验、血清学试验证实其为猪肺炎支原体。该菌株能适应人工培养基的培养,且传代生长良好,液体培养基中培养活菌滴度达109CCU/mL; 菌株有较强的毒力,免疫原性好,可作为疫苗备用菌株,该菌株的分离鉴定为研制猪支原体肺炎灭活疫苗奠定了基础。

Abstract:
Twelve lung samples from pigs with suspected infection by Mycoplasma hyopneumoniae were collected from some pig farms in the country. The strain was further identified to be Mycoplasma hyopneumoniae by PCR,multiple PCR,and the detection of only the infection of Mycoplasma hyopneumoniae were collected for isolation by the method of shearing, samples were cultured and purified in liquid medium.According to the results of sequencing,morphological observation, biochemical characteristics detection and serological test,The isolate strain was identified as Mycoplasma hyopneumoniae. The strain can adapt to the culture of artificial synthetic medium,viable count reached 109CCU/mL. The animal test showed that the isolated strain could cause typical clinical syndrome of swine mycoplasma hyopneumoniae;immunogenicity test showed that the strains had good immunogenicity,and provided the necessary condition for Mycoplasmal pneumonia vaccine research.
