沼泽红假单胞菌发酵提高黄芪多糖含量的条件优化

摘要:
利用沼泽红假单胞菌发酵黄芪,研究发酵对黄芪多糖含量的影响。在单因素试验的基础上,以发酵时间(A)、菌种接种量(B)、发酵温度(C)和初始培养基pH值(D)为4个因素,每个因素设3水平,采用L9(34)正交表,以黄芪多糖含量为指标进行正交试验优化发酵条件。依据F值水平将各因子对黄芪多糖含量的影响按主次顺序排列为:发酵时间>发酵温度>菌种接种量>初始培养基pH值;最佳发酵条件为A3B3C3D1,即在初始培养基pH为6.2,菌种接种量10%和发酵温度40℃的条件下发酵时间6d,黄芪多糖的含量可高达111.40μg/mL,与对照组相比含量提高了51.56%。研究结果表明,利用沼泽红假单胞菌发酵黄芪提高了黄芪多糖的含量,这为提高黄芪资源的利用率提供参考。

Optimization of Conditions for Increasing the Contents of Astragalus Polysaccharide by Microbial Fermentation

Abstract:
In order to investigate the effect on the content of Astragalus polysaccharide from Astragalus fermented by Rhodopseudomonas palustris. The L9(34) orthogonal table and content of Astragalus polysaccharide were employed to select the optimum technique by inoculated quantity, fermentation temperature, initial pH of medium and fermentation time as the four main factors, and three levels were adopted for each factor. The results showed that the optimal technology of Rhodopseudomonas palustris fermented Astagulus were as follows: fermentation time of 6d, fermentation temperature of 40℃, inoculated quantity of 10%, initial pH of 6.2; Under this condition, the content of Astragalus polysaccharide in fermentation medium is 111.40μg/mL. The content of Astragalus polysaccharide in fermentation group was increased by 51.56% than that in the control group. This study provides a reference to optimize the process condition of the Rhodopseudomonas palustris fermented Astragalus, which is of great significance for the efficient use of Astragalus.
