猪瘟病毒片状载体培养生产工艺研究

摘要:
为提高猪瘟抗原的病毒含量,减少猪瘟疫苗的副反应,降低生产成本,对猪瘟病毒片状载体培养生产工艺进行了研究。将ST细胞接种到片状载体上培养,设计了最佳细胞接种密度、接毒量、一收时间、二收时间、连续收获间隔时间、连续收获次数、成品免疫效果的试验方法,确定了最佳生产工艺。结果表明,按照0.5×106/mL的密度接种片状载体培养,72 h后弃去原培养液,加入含7%猪瘟种毒的细胞维持液继续培养,每72 h收获换液一次,可连续收获6次以上,猪瘟抗原的病毒含量能达到1×107 RID/mL,比传统转瓶生产工艺提高了10~20倍,且制备的成品免疫效果良好。本研究成功建立了猪瘟病毒片状载体培养的最佳生产工艺,验证了片状载体培养抗原的免疫效果,为高品质猪瘟疫苗的生产和研究提供了新思路。

Abstract:
To enhance the viral content of classical swine fever antigen, mitigate the side effects associated with classical swine fever vaccine, and reduce production costs, we investigated the production process of classical swine fever virus flake carrier culture. ST cells were inoculated onto flake carriers for cultivation, and a series of experimental methods were employed to optimize parameters, including cell inoculation density, toxic dose, initial and subsequent harvest time, continuous harvest interval time, number of consecutive harvests, and immune efficacy of the final product. Subsequently, the optimal production process was determined. The results revealed that the culture was carried out with a density of 0.5×106/mL on flake carriers. After 72 hours, the original culture medium was replaced with a cell maintenance medium containing 7% classical swine fever virus to sustain the culture. Medium replacement and harvest were performed every 72 hours, resulting in a virus content of classical swine fever antigen reaching 1×107 RID/mL, a significant increase compared to the traditional bottle transfer production process by 10 to 20 times. The prepared product exhibited excellent immune effects. This study successfully established the optimized production process for the cultivation of classical swine fever virus in flake carriers, verified the immune effect of antigen cultivation on such carriers, and provided new ideas for the production and research of high-quality classical swine fever vaccines.
