高效液相色谱法测定大承气散中大黄酚、大黄酸、大黄素、芦荟大黄素、大黄素甲醚的含量

摘要:
本实验建立了采用高效液相色谱技术(HPLC)测定大承气散中大黄酚、大黄酸、大黄素、芦荟大黄素、大黄素甲醚的含量。采用HypersilGOLD (250mm×4.6mm,5um),以0.1%磷酸溶液为流动相A、以乙腈为流动相B进行梯度洗脱,流速1.0mL/min,检测波长为225nm,柱温为40℃,进样体积为20ul。大黄酚在10.0~100.0μg范围内线性关系良好,平均回收率为101.4%(RSD=1.5%);大黄酸在10.5~105.0μg范围内线性关系良好,平均回收率为98.1%(RSD=1.7%);大黄素在11.0~110.0μg范围内线性关系良好,平均回收率为99.4%(RSD=2.1%);芦荟大黄素在14.0~140.0μg范围内线性关系良好,平均回收率为99.8%(RSD=2.1%);大黄素甲醚在15.7~157μg范围内线性关系良好,平均回收率为98.4%(RSD=1.7%)。结果表明该方法有较好的分离度、准确度,可用于测定大承气散中大黄酚、大黄酸、大黄素、芦荟大黄素、大黄素甲醚的含量,为大承气散的质量评价提供数据支持。

Abstract:
Abstract:This experiment established a high-performance liquid chromatography (HPLC) technique to determine the content of chrysophanol,rhein,emodin,aloe-emodin and physcion in Dachengqisan.The sample was separated on a Hypersil GOLD (250mm×4.6mm,5um)with acetonitrile-0.1% phosphoric acid in gradient elution.The flow rate was 1.0 mL/min,The detection wavelength was 225nm,and the column temperature was 40℃,the injection volume was 20ul. The linear relationship of chrysophanol within the range of 10.0 to 100.0ug is good, with an average recovery rate of 101.4% (RSD=1.5%); The linear range of rhein was 10.5 to 105.0ug, with an average recovery rate was 98.1% (RSD=1.7%); The linear range of emodin within was 11.0 to 110.0ug, with an average recovery rate was 99.4% (RSD=2.1%);The linear range of aloe-emodin was 14.0 to 140.0ug,with an average recovery rate was 99.8% (RSD=2.1%); The linear range of physcion was 15.7 to 157.0ug,with an average recovery rate was 98.4% (RSD=1.7%). The results showed that the method has good separation and accuracy, and can be used to determine the content of chrysophanol,rhein,emodin,aloe-emodin and physcion in Dachengqisan, and provide data support for the quality evaluation of Dachengqisan.
