甘草提取物对左氧氟沙星在大鼠体内药动学及组织分布的影响

摘要:
研究甘草提取物对左氧氟沙星在大鼠体内的药动学及组织分布的影响。将SD大鼠随机分成单用组和联用组,单用组和联用组分别予0.9%氯化钠溶液和甘草提取物(0.5 g.kg -1,qd×7 d),灌胃给予左氧沙氟星(20 mg.kg -1)后按时间点连续采样, 采用HPLC法测定给药后血浆以及组织中的左氧氟沙星浓度,数据采用DAS2.0进行分析。结果:与单用组相比,联用组的左氧氟沙星主要药动学参数tmax、t1/2、MRT0-36h、MRT0-∞均显著增大(P<0.05);左氧氟沙星在各组织中浓度水平,呈现出先降低后增加的特点,但分布特征没有改变。结论:甘草提取物能减慢左氧氟沙星在体内的吸收和消除速度,促进其向组织中转运和分布,延长药物在体内的作用时间。

Effect of Licorice Extract on Pharmacokinetics and Tissue Distribution ofLevofloxacin in Rats

Abstract:
To study the effect of licorice extract on the pharmacokinetics and tissue distribution of levofloxacin in rats. The SD rats were randomly divided into the single group and the combined group, that were administrated respectively with normal saline and extract of licorice. After pretreated with extract of licorice (0.5 g?kg-1,qd) for 7 d, levofloxacin was given to the rats by intragastric administration at the dose of 20 mg?kg-1. The concentrations of levofloxacin in plasma and tissue were determined by HPLC. Pharmacokinetic data were calculated by DAS2.0. Comparision with the single administration group, the main pharmacokinetic parameters (t1/2, tmax, MRT0-36h, MRT0-∞) were significantly increased (P<0.01), the concentration level of levofloxacin in tissuet showed a feature of first decreasing followed by increasing, but the characteristic of tissue distribution was not changed. The metabolic characteristics of levofloxacin were influenced by licorice extract in rats, including decreasing the speed of absorption, retarding the speed of elimination, accelerating the distribution from blood to tissue, lengthening the time of drug action.
